Is there a requirement for mounted threaded rods to the ceiling to be fire caulked around the mounting points?

It is technically outside the scope of the firestop manufacturer to determine if you need to firestop an opening. As such, I would reach out to your Authority Having Jurisdiction or local code official to confirm whether or not firestopping is required.
However, based on your brief description, this is an application that we have addressed before. The only system we have that is somewhat close is F-C-7042, but I do not believe it is an exact match. As such, we can address your application through an Engineering Judgment (EJ), which will be a custom detail showing your exact application with the firestop components required to maintain the stated ratings.
